Em 1651 Giambattista Pamphilj, depois de ascender ao trono papal com o nome de Inocêncio Décimo (1644-1655), estabeleceu a Galeria.
Inclui pinturas de Rafael, Ticiano, Domenichino, Parmigianino, Caravaggio, Annibale e Ludovico Carracci, entre outros.
